Nanocomposite of multiwalled-carbon-nanotubes (MWCNT) has been homogeneously integrated (0 – 9 wt%) with polymer, poly (vinylidene fluoride, PVDF) to yield 300 micron film. The PVDF + 9 wt% MWCNT sample of density 1.41 g/cm3 show specific shielding effectiveness (SSE) of 17.7 dB/(g/cm3) (99.6% EMS), with maintained hardness and improved conductivity. With multilayer stacking (900 microns) of these films of density 1.37 g/cm3, the sample showed increase in SSE to 23.3 dB/(g/cm3) (99.93% EMS). Uniform dispersion of MWCNTs in the PVDF matrix gives rise to increased conductivity in the sample beyond 5 wt% MWCNT reinforcement. Neurodegenerative patients face early mitochondrial dysfunction, impacting energy utilization. Amyloid-ß and tau problems both have an effect on mitochondria, which leads to mitochondrial malfunction and, ultimately, the onset of Alzheimer's disease. Cellular oxygen interaction yields reactive oxygen species within mitochondria, initiating oxidative damage to mitochondrial constituents. Parkinson's disease, linked to oxidative stress, α-synuclein aggregation, and inflammation, results from reduced brain mitochondria activity. Mitochondrial dynamics profoundly influence cellular apoptosis via distinct causative mechanisms. The condition known as Huntington's disease is characterized by an expansion of polyglutamine, primarily impacting the cerebral cortex and striatum. Research has identified mitochondrial failure as an early pathogenic mechanism contributing to HD's selective neurodegeneration. The mitochondria are organelles that exhibit dynamism by undergoing fragmentation and fusion processes to attain optimal bioenergetic efficiency. They can also be transported along microtubules and regulate intracellular calcium homeostasis through their interaction with the endoplasmic reticulum. Additionally, the mitochondria produce free radicals. The functions of eukaryotic cells, particularly in neurons, have significantly deviated from the traditionally assigned role of cellular energy production. Most of them are impaired in HD, which may lead to neuronal dysfunction before symptoms manifest. This article summarizes the most important changes in mitochondrial dynamics that come from neurodegenerative diseases including Alzheimer's, Parkinson's, Huntington's and Amyotrophic Lateral Sclerosis. Finally, we discussed about novel techniques that can potentially treat mitochondrial malfunction and oxidative stress in four most dominating neuro disorders.
Development and Validation of Stability-Indicating HPTLC Method for Estimation of Secnidazole in Bulk Drug and Pharmaceutical Dosage Form
A simple, selective, precise and stability-indicating high-performance thin layer chromatography ... more A simple, selective, precise and stability-indicating high-performance thin layer chromatography (HPTLC) method for the analysis of Secnidazole both in bulk drug and pharmaceutical formulation has been developed and validated. The method employed HPTLC aluminium plates precoated with silica gel 60 F254 as the stationary phase. The solvent system consisted of Toluene: methanol: triethylamine (4: 1: 0.4 v/v/v). The system was found to give compact spot for Secnidazole (Rf value of 0.50 ± 0.03). Densitometric analysis of Secnidazole was carried out in the absorbance mode at 313 nm. The linear regression analysis data for the calibration plots showed good linear relationship with r 2 = 0.999 ± 0.0017 with respect to peak area in the concentration range 200–1200 ng per spot. The mean values ± SD of slope and intercept were 10.081 ± 1.56 and 3607.5 ± 1.48, respectively, with respect to peak area. The method was validated for precision, recovery and robustness. Since they are poisonous, they are processed with various media like Godugdha (cow's milk), Kanji (sour gruel), Nimbu Swarasa (Lemon Juice), etc. before being incorporated into the Ayurvedic formulations. This study investigates the use of Gomutra (Cow Urine), normal saline (NaCl), and water as detoxification media. A stainless-steel vessel with a dolayantra was used to immerse the seeds for 3 hours of boiling during the detoxification process. Detoxification using Gomutra gave a reddish tinge to the seeds compared to the light brown color seen with NaCl medium. The physicochemical changes in detoxification that occurred in the gomutra treated gunja seeds were validated by TLC and Ex Vivo investigations (agglutination assay). To characterize the extract using TLC, the mobile phase was Toulene: ethyl acetate: glacial acetic acid (6.5: 3.3: 0.2). The removal of distinctive bands in the TLC profile of processed gunja seeds suggested that the gunja seed extract's composition had changed.
